Sve više i više ljudi diljem svijeta pati jer njihov imuni sustav više ne može razlikovati zdrave stanice od napadačkih mikroorganizama. Obrana od bolesti koja ih je nekada štitila umjesto toga napada njihova tkiva i organe. Ulažu se veliki međunarodni istraživački napori u borbi protiv ovog trenda – uključujući inicijativu na londonskom Institutu Francis Crick, gdje su dva svjetska stručnjaka, James Lee i Carola Vinuesa, osnovali zasebne istraživačke grupe kako bi pomogle u utvrđivanju točnih uzroka autoimunih bolesti, jer uvjeti su poznati.

“Broj autoimunih slučajeva počeo je rasti prije otprilike 40 godina na zapadu”, rekao je Lee za Observer. “Međutim, sada vidimo da se neki pojavljuju u zemljama koje nikada prije nisu imale takve bolesti. Autoimune bolesti se kreću od dijabetesa tipa 1 do reumatoidnog artritisa, upalne bolesti crijeva i multiple skleroze. U svakom slučaju, imunološki sustav doživljava kratki spoj i napada zdravo tkivo umjesto infektivnih agenasa. Samo u Ujedinjenom Kraljevstvu, najmanje 4 milijuna ljudi razvilo je takva stanja, a neki pojedinci pate i više od jednog. Na međunarodnoj razini, sada se procjenjuje da slučajevi autoimunih bolesti rastu između 3 posto i 9 posto godišnje. Većina znanstvenika vjeruje da čimbenici okoliša igraju ključnu ulogu u ovom porastu.

"Ljudska genetika nije se promijenila u posljednjih nekoliko desetljeća", rekao je Lee, koji je prije bio sa sjedištem na Sveučilištu Cambridge. “Dakle, nešto se mora mijenjati u vanjskom svijetu na način koji povećava našu predispoziciju za autoimune bolesti.” Ovu ideju podržala je Vinuesa, koji je prije bio smješten na Australskom nacionalnom sveučilištu. Oba su znanstvenika naglasila da je individualna osjetljivost uključena u zarazu takvim bolestima koje također uključuju celijakiju, kao i lupus, koji izaziva upalu i oticanje te može uzrokovati oštećenje različitih organa, uključujući srce. “Ako nemate određenu genetsku osjetljivost, nećete nužno dobiti autoimunu bolest, bez obzira koliko Big Maca jedete”, rekao je Vinuesa. “Ne možemo puno učiniti da zaustavimo globalno širenje franšiza brze hrane. Umjesto toga, pokušavamo razumjeti temeljne genetske mehanizme koji podupiru autoimune bolesti i čine neke ljude osjetljivima, ali druge ne. Želimo se pozabaviti tim pitanjem na toj razini.”

Takav rad leži u srži Leejevih i Vinuesinih napora, čiji je cilj otkriti kako ti različiti genetski putovi djeluju i razotkriti mnoge različite vrste bolesti koje liječnici sada promatraju. Sve više i više ljudi diljem svijeta pati jer njihov imuni sustav više ne može razlikovati zdrave stanice od napadačkih mikroorganizama. Obrana od bolesti koja ih je nekada štitila umjesto toga napada njihova tkiva i organe. Ulažu se veliki međunarodni istraživački napori u borbi protiv ovog trenda – uključujući inicijativu na londonskom Institutu Francis Crick, gdje su dva svjetska stručnjaka, James Lee i Carola Vinuesa, osnovali zasebne istraživačke grupe kako bi pomogle u utvrđivanju točnih uzroka autoimunih bolesti, jer uvjeti su poznati.

“Broj autoimunih slučajeva počeo je rasti prije otprilike 40 godina na zapadu”, rekao je Lee za Observer. “Međutim, sada vidimo da se neki pojavljuju u zemljama koje nikada prije nisu imale takve bolesti. Na primjer, najveći nedavni porast slučajeva upalnih bolesti crijeva zabilježen je na Bliskom istoku i u istočnoj Aziji. Prije toga, te države gotovo da nikad nisu vidjele tu bolest.”

Autoimune bolesti se kreću od dijabetesa tipa 1 do reumatoidnog artritisa, upalne bolesti crijeva i multiple skleroze. U svakom slučaju, imunološki sustav doživljava kratki spoj i napada zdravo tkivo umjesto infektivnih agenasa. Samo u Ujedinjenom Kraljevstvu, najmanje 4 milijuna ljudi razvilo je takva stanja, a neki pojedinci pate i više od jednog. Na međunarodnoj razini, sada se procjenjuje da slučajevi autoimunih bolesti rastu između 3 posto i 9 posto godišnje. Većina znanstvenika vjeruje da čimbenici okoliša igraju ključnu ulogu u ovom porastu.

"Ljudska genetika nije se promijenila u posljednjih nekoliko desetljeća", rekao je Lee, koji je prije bio sa sjedištem na Sveučilištu Cambridge. “Dakle, nešto se mora mijenjati u vanjskom svijetu na način koji povećava našu predispoziciju za autoimune bolesti.” Ovu ideju podržala je Vinuesa, koji je prije bio smješten na Australskom nacionalnom sveučilištu. Ukazala je na promjene u prehrani koje su se događale jer je sve više zemalja usvajalo zapadnjačku prehranu i ljudi kupovali više brze hrane.

"U prehrani s brzom hranom nedostaju određeni važni sastojci, poput vlakana, a dokazi sugeriraju da ova promjena utječe na mikrobiom osobe - zbir mikroorganizama koje imamo u crijevima i koji igraju ključnu ulogu u kontroli različitih tjelesnih funkcija. Ove promjene u našim mikrobiomima tada pokreću autoimune bolesti, kojih je sada otkriveno više od 100 vrsta", rekla je Vinuesa.

Oba su znanstvenika naglasila da je individualna osjetljivost uključena u zarazu takvim bolestima koje također uključuju celijakiju, kao i lupus, koji izaziva upalu i oticanje te može uzrokovati oštećenje različitih organa, uključujući srce. “Ako nemate određenu genetsku osjetljivost, nećete nužno dobiti autoimunu bolest, bez obzira koliko Big Maca jedete”, rekao je Vinuesa. “Ne možemo puno učiniti da zaustavimo globalno širenje franšiza brze hrane. Umjesto toga, pokušavamo razumjeti temeljne genetske mehanizme koji podupiru autoimune bolesti i čine neke ljude osjetljivima, ali druge ne. Želimo se pozabaviti tim pitanjem na toj razini.”

Ovaj zadatak je moguć zahvaljujući razvoju tehnika koje sada omogućuju znanstvenicima da uoče male DNK razlike među velikim brojem pojedinaca. Na taj način moguće je identificirati uobičajene genetske obrasce među onima koji boluju od autoimunih bolesti. "Do nedavno, jednostavno nismo imali alate za to, ali sada imamo nevjerojatnu moć sekvenciranja DNK u velikim razmjerima i to je sve promijenilo", rekao je Lee. “Kad sam počeo istraživati, znali smo oko pola tuceta varijanti DNK koje su bile uključene u pokretanje upalne bolesti crijeva. Sada znamo za više od 250.”

Takav rad leži u srži Leejevih i Vinuesinih napora, čiji je cilj otkriti kako ti različiti genetski putovi djeluju i razotkriti mnoge različite vrste bolesti koje liječnici sada promatraju. "Ako pogledate neke autoimune bolesti - na primjer, lupus - nedavno je postalo jasno da postoji mnogo različitih verzija njih, koje mogu biti uzrokovane različitim genetskim putevima", objasnila je Vinuesa pa nsatavila: “A to ima posljedicu kada pokušavate pronaći pravi tretman. Lee je također naglasio da sve veći broj slučajeva autoimunih bolesti diljem svijeta znači da su novi tretmani i lijekovi sada hitno potrebniji nego ikada prije. “Trenutno ne postoje lijekovi za autoimune bolesti, koje se obično razvijaju kod mladih ljudi – dok pokušavaju završiti školovanje, dobiti prvi posao i imati obitelji.
